Read this and loved it for a couple of reasons! First is that we're approaching this kind of thing on our site at harmoniker.com. Less of the "follow" and "friend list" stuff and more of the "direct connection between people talking about interesting things".
Secondly, we're big WOW fans (you've heard of it, right? ;-) and have just revised our "/Games/World Of Warcraft" categories to give players a place to get together outside the game. Good place to plug in a few "external" interests too and find similar people. It's just started but we're hoping to build it quickly.
Hope you think it's all a step forward!

@Noah
Clickable hashtags have the disadvantage of only telling you about it if someone you follow uses it. The point of "following" a conversation would be to hear from it even if your friends don't mention it.
I guess you could have a list of hashtags "buttons" somewhere in a corner and click on them when you feel like it, but this solution doesn't provide the "free flow feed" that is so enjoyable in twitter.
And yes, I love alliterations.

@David
Well, let's just say this would take us down a dangerous path of "feature adding". Separate hashtags is cool, but why create fields for URLs, pictures and locations too? It's an appealing idea and I would actually be for it, but let's not take it too far and risk the sacro-saint simplicity.
Besides, a hashtag isn't too much of a problem either; if you can fit a tinyURL into your message you should be able to fit a hash tag too, right?

On the comment of hashtags, i agree with you. One thing i would add to this is Twitter should seperate the tags from the tweet like Flickr tags or a delicious bookmark. This would allow twitter users to make more use of hash tags without sacrificing their 140 characters.

Or, instead of following #hashtags, just make them clickable and have it take you to the search page w/ the hashtag entered as the search query.
